In Vitro Susceptibility of Cryptococcus neoformans to Amphotericin B, Nystatin, 5-fluorocytosine, Ketoconazole, Mconazole, and Rifampicin Alone and in Combination


Abstract
In vitro antifungal activities of ketonazole, miconazole, amphotericin B, nystatin, 5-fluorocytosine and rifampicin in combined with ketoconazole/miconazole are determined against 4 strains of Cryptococcus enoformans.
The minimum inhibitory concentrations of ketoconazole, miconazole, amphotericin B, nystatin, 5-fluorocytosine and rifampicin for tested strains were ranged from 0.047pg m1-1 - 0.094 pg ml-¢¥, 0.78 pg m1 1 - 1.50 pg ml-¢¥, 0.39 pg m1¢¥1 - 1.56 pg ml-1, 40.0 pg ml-1 - 50.0 pg ml-1, 0.25 pg ml-1 - 6.25 pg ml-1 and > 100.0 pg ml-1.
The growth curve experiment with on strain of Qyptococcus neoformans. WCM-1 demonstrated that the combination of ketoconazole/miconazole produced a decrease in number of colony forming unit of <2 logs in 24 hours of observation.
On the basis of theses studies, it is concluded that ketoconazole/miconzaole are synergistic against Cryptococcus neoformans under experimental conditions.
